When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

    Involved in cytoskeletal rearrangements required for phagocytosis of apoptotic cells and cell motility. Functions as a guanine nucleotide exchange factor (GEF), which activates Rac Rho small GTPases by exchanging bound GDP for free GTP. Its GEF activity may be enhanced by ELMO1.
    Tissue specificity: Highly expressed in placenta, lung, kidney, pancreas and ovary. Expressed at intermediate level in thymus, testes and colon.

    Function:

                                        乳腺癌轉移的新標記物DOCK1
        乳腺癌是一種常常發(fā)生轉移的癌癥。將近90%的乳腺癌患者都因為癌癥擴散而死,因此,理解這一危險過(guò)程的細胞和分子基礎機制是十分重要的。
        近日,加拿大蒙特利爾臨床研究所(Institut de recherches cliniques de Montreal, IRCM)的科學(xué)家在乳腺癌研究方面取得了一個(gè)重要的發(fā)現,他們確定了,一種名為DOCK1的蛋白質(zhì)可以作為潛在的靶標用于減少乳腺癌患者的癌轉移。相關(guān)研究結果刊登在了近期出版的《PNAS》上。
        研究人員在乳腺癌治療方面盡管已經(jīng)取得了重要的進(jìn)展,但是對于癌癥擴散的機制卻知之甚少??茖W(xué)家正在試圖確定能夠調節癌轉移過(guò)程的蛋白質(zhì),因為它們或許可以作為新的藥物靶標,并與現有的方法相結合促進(jìn)治療的效果。
        在這項研究中,2個(gè)主要的癌癥亞型——HER-2+型和Basal-like型,很容易轉移和復發(fā),并最終導致患者的低生存率。該調查以HER-2+(人類(lèi)表皮生長(cháng)因子受體2)型為研究對象, 這代表了大約25%的乳腺癌病例。 HER2陽(yáng)性腫瘤比其它類(lèi)型的腫瘤更傾向于快速的發(fā)展和擴散。
        研究人員解釋到,通過(guò)研究HER2+乳腺癌小鼠模型,確定了DOCK1蛋白質(zhì)是癌轉移的一個(gè)重要的調控因子。當去除了小鼠中的這一蛋白質(zhì)后,乳腺癌的肺轉移大大減少了。此外,研究發(fā)現DOCK1蛋白質(zhì)有助于腫瘤的生長(cháng)。
        為了證明DOCK1的表達與乳腺癌預后之間的關(guān)系,研究人員對包含患者基因信息的幾個(gè)數據庫進(jìn)行了分析。結果發(fā)現,在HER-2+型或Basal-like型乳腺癌中,高水平的DOCK1與較差的預后以及疾病的復發(fā)有關(guān)。
